I just downloaded the Jamstix demo I got everything up and running but I can not seem to get the audio jam to work. The red audio button lights up when I play but how do you get the drums to play along and improvise over what you are playing? This is the main reason why I downloaded it and can not figure it out. Can someone please help? I am using Reaper by the way. Thanks!!!

Jamstix does not improvise over your audio signal. That is not possible in real-time with the data that can be gleaned from an audio signal.

Instead, you select style, drummer and kit you'd like to use, let the host play and JS will then use the volume data from the audio signal to adjust dynamics and play rules. For example, it will switch to cross-sticking on low quiet passages and ride cymbal on loud ones.
